Hepatic Microsomal Oxidative Metabolism of Pesticides and Other Xenobiotics in Pregnant CD, Mice

چکیده

Pregnancy-related changes in oxidative metabolism of several xenobiotics including pesticides were examined in the hepatic microsomes of CD, mice. The effect of pregnancy on hepatic microsomal cytochrome P-450~catalyzed substrate oxidation was found to be dependent upon the type of reaction examined. Not all substrates undergoing the same reaction showed identical changes during pregnancy. Those enzyme activities which exhibited a decline in specific activity during pregnancy generally exhibited no change in total hepatic capacity. Enzymes posting no change in specific activity throughout gestation generally showed large increases in total hepatic activity. Phorate S-oxidation was catalyzed by both microsomal flavin-containing monooxygenase (MFMO) and cytochrome P-450. Moreover, there was no pregnancy-related change in either MFMO or total enzymatic (MFMO plus cytochrome P-450) phorate S-oxidation. 6 1985 Academic Press. Inc.

Oxidative metabolism of xenobiotics during pregnancy: significance of microsomal flavin-containing monooxygenase.

Pregnancy related changes in oxidative metabolism of model substrates were examined in CD1 mice. As compared to nonpregnant females, a significant decrease in the hepatic microsomal aminopyrine-but not in dimethylaniline-N-demethylase activity was observed in pregnant mice. The rates of microsomal flavin-containing monooxygenase-catalyzed N-oxidation of dimethylaniline remained relatively uncha...

An Investigation of the Role of Microsomal Oxidative Metabolism in the in Vivo Genotoxicity of 1,2-Dichloroethane. STORER, R. D.. AND CONOLLY, R. B. (1985). Toxicol. Appl. Pharmacol. 77,36-46. In vitro studies have demonstrated that two different metabolic pathways, glutathione conjugation mediated by the glutathione S-transferases and microsomal oxidation, may be involved in the genotoxicity a...

Inactivation of the hepatic cytochrome P450 system by conditional deletion of hepatic cytochrome P450 reductase.

Cytochrome P450 (CYP) monooxygenases catalyze the oxidation of a large number of endogenous compounds and the majority of ingested environmental chemicals, leading to their elimination and often to their metabolic activation to toxic products. This enzyme system therefore provides our primary defense against xenobiotics and is a major determinant in the therapeutic efficacy of pharmacological a...

MFÔs are a large diverse superfamily of enzymes found in all insect tissues. They are involved in the metabolism of xenobiotics (e.g. drugs, pesticides and plant toxins) and endogenous compounds (e.g. ecdysteroids and juvenile hormones). They are also involved in bioactivation of phosphorothioate compounds such as organophosphorus insecticides. They have very diverse activities like hydroxyla...

The aim of this study was to further characterize the expression and function of human CYP2B6 in a recently generated CYP2A13/ 2B6/2F1-transgenic (TG) mouse model, in which CYP2B6 is expressed selectively in the liver. The inducibility of CYP2B6 by phenobarbital (PB) and dexamethasone (DEX), known inducers of CYP2B6 in human liver, was examined in the TG mice, as well as in TG/Cyp2abfgs-null (o...
